Available Assays

ADC Plasma Stability Assay

Incubate your ADC in human or mouse plasma and monitor payload release over time by LC-MS. Provides critical in vitro PK data before committing to in vivo studies.

Lysosomal Release Assay

Cell-based assay confirming that your cleavable linker releases payload in the lysosomal compartment after internalization. Key mechanistic validation for pH-sensitive or cathepsin-cleavable linkers.

Kupffer Cell Uptake Assay

In vitro assessment of ADC uptake by liver-resident macrophages (Kupffer cells). Provides early insight into clearance risk, Fc-mediated interactions, and potential impact on pharmacokinetics.

Target Binding Confirmation

ELISA and/or flow cytometry to confirm that conjugation has not impaired antigen or target cell binding affinity or specificity. Run pre- and post-conjugation for direct comparison.

ADC Internalization Assays

Quantify receptor-mediated internalization kinetics in antigen-positive and antigen-negative cell lines, via flow cytometry.

Evaluation of Effector Function

Evaluate ADCC, ADCP, and CDC activity of your ADC or unconjugated antibody using primary human immune effector cells or complement, alongside direct Fc receptor binding. Critical for understanding Fc-mediated and non-cytotoxic mechanisms of action.

Direct Cytotoxicity Assays

Dose-response cytotoxicity in antigen-positive and antigen-negative cell lines to determine IC50, maximum killing, and on-target selectivity of your ADC.

Bystander Effect Co-Culture Assays

Evaluate bystander killing of antigen-negative cells in co-culture with antigen-positive target cells. Particularly relevant for membrane-permeable payloads and heterogeneous tumor models.
