C1q and Complement

C1q is the initiating component of the classical pathway of the complement system, a key part of the immune response. C1q recognizes and binds to the Fc region of antibody-antigen complexes, as well as directly to certain pathogens, activating the complement cascade. This activation leads to C3 convertase formation, driving inflammation, opsonization (marking pathogens for destruction), and the assembly of the membrane attack complex (MAC), which can directly lyse target cells. Depending your therapeutic’s mechanism of action and target disease, it may be critical to confirm C1q recognition via binding or demonstrate C1q evasion for safety assessment.

The AlphaLISA method for detecting human C1q binding to the antibody Fc region utilizes a biotinylated anti-human IgG Fab antibody, which binds to Streptavidin Donor beads. These beads capture and aggregate the IgG antibody present in the sample. Simultaneously, AlphaLISA anti-C1q Acceptor beads bind C1q protein via its collagen-like domain. When the antibody binds to C1q, the Donor and Acceptor beads are brought into proximity. Upon excitation of the Donor beads, they generate singlet oxygen, initiating an energy transfer cascade within the Acceptor beads. This results in a distinct light emission peak at 615 nm.
