T cell-Dependent Cellular Cytotoxicity (TDCC) Assay

T-cell Engagers (TCEs) have gained popularity in the field of immunotherapy due to their ability to dually interact with two antigens of interest, often redirecting T cell effector function via CD3 toward cancer cells expressing a tumor antigen.
At iQ Biosciences, our T cell-dependent cellular cytotoxicity (TDCC) assay can help you screen candidate TCEs, including bispecific antibodies (e.g., BiTEs, DARTs, and TandAbs), TCR-based therapeutics, and other T-cell-redirecting biologics,, for effective cytotoxicity against target cell(s) of interest. Target cells, typically cancer cell lines, are labeled with a cell trace dye and then co-cultured Pan T (CD3+) cells previously isolated from whole blood obtained from healthy donors. Assays are performed at defined effector:target (E:T) ratios and timepoints, depending on relevant factors. A range of therapeutic concentrations are tested to elucidate any dose-dependent cytotoxicity. We also provide or recommend appropriate positive controls (recombinant versions of FDA-approved BiTE antibodies, for example, as available) and negative controls to ensure assay validity and accuracy. After incubating the co-culture with therapeutic molecules or controls, viability and/or depletion of target cells is determined by flow cytometric or plate-reader based analysis.
This assay can be combined with cytokine quantitation or T cell activation marker analysis, to provide additional functional insights. In addition to Human T cell TDCC assays, we offer Cynomolgus Monkey (cyno) TDCC assays.
